Having recently defeated him in battle, Ka-Zar takes the unconscious Spider-Man away to learn more about him. Ka-Zar explains that he had been convinced to capture Spider-Man by J. Jonah Jameson. Spider-Man also realizes that he has now shaken his amnesia following their battle.

J. Jonah Jameson receives a call from Spencer Smythe who claims to have a new plan to finish Spider-Man. John Jameson and Captain Stacy discuss their recent attempts at clearing Spider-Man’s name. At the hospital, Gwen Stacy and Harry Osborne visit Aunt May who continues to call for her nephew Peter.

Spider-Man explains his recent memory loss and how he was tricked into thinking he was the partner of Doctor Octopus and Ka-Zar believes him, but suddenly leaps from the top of the building to help Zabu who is being tracked my police. Spider-Man remembers that his Aunt is in the hospital and heads back to his apartment to get some clothes.

Jonah pays Smythe a visit and is shown his latest invention, the Spider-Slayer. It is larger and more powerful than the previous mechanism. The new robot is also twice as sensitive as the previous model and will be able to track Spider-Man from a much greater distance.

Spider-Man finally gets back to his apartment, but is surprised by the Spider-Slayer which has managed to track him across the city. It also displays its new weapon, the Destructo-Beam, which fires out at the wall crawler narrowly missing him. Jonah is surprised by the deadly blast and does not approve, but Smythe will not be beaten again no matter what the cost. He pushes Jonah away from the controls and takes over. Spider-Man eventually leads the Spider-Slayer to Smythe’s lab where its circuits overload due to the presence of dozens of spiders Smythe keeps for his experiments.

The Spider-Slayer Mark II is the first to actually be refered to as a "Spider-Slayer".
